Covariate adjustment in clinical trials leads to gains in statistical power. Our simulation study quantifies the impact of covariate adjustment in different disease indications. Adopting an evidence-based approach will significantly improve the success rate of trials. New prognostic modalities such as digital pathology can further de-risk and accelerate clinical trials. Broader eligibility criteria in trials are more attractive once covariate adjustment is optimized.
